"to be supposed to" is an obligation, usually imposed by someone else. Ex: I'm supposed to clean my house (because my mom told me to). It could also be an obligation that you agreed to though, so not necessarily something imposed by someone else. Ex: The party's tomorrow? Oh, sorry, I can't come. I'm supposed to babysit my little niece tomorrow (because I told my sister that I would).

By contrast, "should do" is just a suggestion. It's good advice; kinda like "it would be a good idea to". Ex: I should clean my house (because it's really dirty and it's grossing me out). A lesser-used synonym for "should do" is "ought to do".
