Hello! In detective Akechi Kogoro, Kobayashi Yoshio is known for being stutter-ish, because of what he usually ruins important or tensed moments. There is a dialogue in which I can’t clearly see the “word-play”. Could someone please explain this moment to me?

I don't think there's "word-play" in this sentence.
If I have to say, he reworded 殺った to 殺した because he was in the situation which he shouldn't use colloquial expression. 殺る is colloquial and a little bit rough word.
