Although both make sense.
‘During’ is a time period. Because summer is a time period, you would say ‘during his summer vacation.’
‘In’ is more of being inside a place, summer isn’t a location so it wouldn’t make sense to say ‘in his summer vacation.’ An example of how to use ‘in’ is: “He went in his car.”

They are pretty much the same, but the first sentence is just a little more gramatically correct/ clear!
Either one is fine, but the term "during" makes more sense in this context since you can't be "in" something like a (summer vaction) but you can discribe a time "during" a (summer vacation)!
